This film, which is based on the renowned Professor Layton puzzle games, introduces Layton and his faithful aide Luke to the big screen, allowing fans to experience more of the same on the big screen. At the start of the film, Layton and his family get a letter from the renowned opera singer, Jenice Kaitly, inviting them to attend her most recent performance. Also, she seeks Layton, Luke, and Remi for help in solving a mystery, claiming that her best friend Melina vanished a year ago and reappeared as a seven year old declaring she had discovered the secret to perpetual life. Layton and his trainees are naturally intrigued by the situation, and they decide to investigate it further at the Crown Petone opera theatre. When they come, Ozro Vislar, the pianist, begins to talk about a game in which the winner will receive "eternal life," and the audience gasps. It is up to Layton, Luke, and Remi to figure out what is going on with the Eternal Diva before it is much too late.
